摘要:
[目的] 探究打破西番莲种子休眠以促进种子萌发的最适培养基及萌发方式,加快其实生苗的繁育。[方法] 以黄果西番莲种子为试验材料,研究不同培养基(1/2MS、3/4MS、MS和PNM)、不同剥壳方式(不剥、只裂不剥、半剥和全剥)及不同质量浓度(0、5、10、15、20 mg·mL-1)赤霉素(GA3)浸种对西番莲种子萌发率和萌发速率的影响。[结果] 4种剥壳处理中,西番莲种子全剥壳处理萌发率最高,为73.63%;GA3浸种中,以5 mg·mL-1GA3浸种萌发率(82.22%)最高,GA3浓度过高则抑制种子萌发;4种培养基中,以PNM培养基萌发率最高(68.89%),显著高于其他培养基。[结论] 西番莲种子在5 mg·mL-1 GA3浸种12 h后全剥壳接种于PNM培养基,是其萌发的最佳方法,萌发率达到82.22%。该法有利于西番莲实生苗的快速繁殖。
Abstract:
[Purpose] To explore the best culture medium and the way of germination for breaking the seed dormancy of Passiflora caerulea seeds and accelerating the propagation of seedlings. [Method] The seeds of P.caerulea were used as experimental material. The effects of different culture media (1/2 MS, 3/4 MS, MS and PNM), different deshelling methods (no peeling, just split, semi-peeling, full peeling), and different concentrations (0, 5, 10, 15, 20 mg·mL-1) of GA3 on the germination rate and germination rate of P.caerulea seeds were studied. [Result] Among the deshelling methods, the germination rate of fully peeled P.caerulea seeds was the highest at 73.63%. The highest seed germination rate (82.22%) was observed after soaking in 5 mg·mL-1 GA3, but the germination was somehow inhibited when the GA3 concentration was too high. Among the culture media tested, the PNM media produced the highest seed germination rate of 68.89%, which was significantly higher than the rates fromthe other media (P<0.01). [Conclusion] The best method for germinating P.caerulea seeds was pre-soaking the seeds in 5 mg·mL-1 GA3 for 12 hours, followed by full peeling and inoculating on PNM medium. The germination rate reached 82.22%. This method was beneficial to the rapid propagation of P.caerulea seedlings.
